Background :Ecto-5'-nucleotidase is a 70 kDa glycosyl phosphatidylinositol-anchored, membrane-bound glycoprotein that catalyzes the hydrolysis of extracellular nucleoside monophosphates into bioactive nucleosides. NT5E catalyzes the terminal step of extracellular adenosine formation from adenosine monophosphate, which drives the regulation of extracellular adenosine levels and the downstream activation of the four G protein-coupled adenosine receptors . Binding of hypoxia-inducible factor to the NT5E gene promoter leads to upregulation of NT5E during hypoxia. The biological roles of NT5E include lymphocyte adhesion, fibrosis , and the regulation of nociception.

Western blot analysis of CD73 expression in MDAMB231 (A) whole cell lysates.

Immunohistochemical analysis of CD73 staining in human kidney formalin fixed paraffin embedded tissue section. The section was pre-treated using heat mediated antigen retrieval with sodium citrate buffer (pH 6.0). The section was then incubated with the antibody at room temperature and detected using an HRP conjugated compact polymer system. DAB was used as the chromogen. The section was then counterstained with haematoxylin and mounted with DPX.
